- W2022146964 abstract "Abstract De novo protein synthesis during early embryogenesis of the amphibian Xenopus laevis was investigated at six different stages of development. Isotopically labeled amino acids were microinjected into embryos and the newly synthesized proteins containing this label were analyzed by polyacrylamide gel electrophoresis on the high resolution two-dimensional gel system described by O'Farrell. Complex, but highly reproducible, changes were observed between each of the six consecutive stages analyzed when the proteins were extracted by the modified O'Farrell procedure described. A greater degree of change in the types of proteins synthesized between the cleavage stage and late blastula was observed than has been previously been reported for other developmental systems. Furthermore, a major, nonrandom shift in the kinds of proteins being synthesized occurs between the gastrula and neurula stages of development. The major de novo synthesized postgastrula proteins appear to be restricted to a rather limited pH and apparent molecular weight region of two-dimensional electrophoretic gels. Actin and tubulin are shown to be present as de novo synthesized proteins at all stages of development but their relative rates of synthesis before the gastrula stage appear to be low. It is also noted, however, that the use of a number of different isotopically labeled amino acid precursors may be necessary in an experiment to detect certain proteins unequivocally." @default.
