- W2022153552 abstract "Celastrus paniculatus has been reported as neuroprotective, hypolipidemic and also enhances swimming time but the molecular mechanism of action of swimming endurance has not been reported. Preliminarily we identified the metabolites of C. paniculatus seed extract (CPSE) by GC–MS which revealed the presence of sesquiterpenes and esters. Further we supplemented mice with 50 mg/kg b/wt CPSE and subjected them for forced swimming test with 10% of body weight tagged to the tail. The exercise endurance performance, biochemical mechanism and metabolic regulation of CPSE were investigated. The results showed that CPSE supplementation enhanced the exercise duration up to two-fold and also regulated the serum biochemical markers such as glucose, blood urea nitrogen, creatine kinase, lactate dehydrogenase and tissue biochemical markers such as glycogen, lactic acid, superoxided dismutase, catalase and lipid peroxidation. Exercise induced muscle and DNA damage were protected by CPSE supplementation as analyzed by TEM and comet assay. Further the extract administration regulated the exercise induced expression of metabolic and oxidative stress markers such as AMPK, Glut-4, PDK4, VEGF, PGC-1α, MCT-1, MCT-4, SOD, CAT and HSP-70 as verified by Western blot analysis. Overall the C. paniculatus supplementation enhanced the mice muscle metabolism and showed potent DNA damage inhibitory activity explaining the possible mechanism of exercise endurance and antioxidative defense status of the seed extract." @default.
