- W2022293865 abstract "Summary o 1. A simple spectrophotometric method for quantitation of three abnormal hemoglobin pigments is described. By this method the concentration of two pigments present in the same blood as met- and carboxyhemoglobin or met- and sulphemoglobin can be measured. The multicomponent system is reduced in each case to a two-component system by means of chemical transformations and isobestic points. 2. The technique is carried out by recording a spectral curve of diluted hemolysed red cells, in which the reduced hemoglobin is converted to oxyhemoglobin. Any elevation of absorbance at 620-630 nm indicates the presence of sulph- and/or methemoglobin which can then be differentiated by addition of cyanide. 3. Quantitation of the abnormal hemoglobin pigments is based on the ratioof the absorbances measured at two wavelengths. The percentages of the abnormal pigments are obtained directly from a graph. 4. The choice of selected wavelengths and the accuracy and sensitivity of themethod is discussed." @default.
