- W2022510931 abstract "Our goal was to detect ovarian tumor associated neo-angiogenesis (TAN) by Doppler ultrasound (DUS) at early stage of OVCA in laying hens, a spontaneous model of human OVCA. Healthy White Leghorn hens (Gallus domesticus) with low egg laying were selected by transvaginal DUS scanning (Z-one, Zonare Medical Systems). Hens were reared under standard conditions. Hen ovaries were scanned for Gray scale evaluation and Doppler indices (resistive index, RI = [systolic velocity diastolic velocity]/systolic velocity and pulsatility index, PI = [systolic velocity − diastolic velocity]/mean) were calculated. Hens were scanned at 15 wk intervals and sacrificed when morphological or vascular abnormalities were suspected by ultrasound scanning or at the end of the study (45 weeks). Diagnosis were made at sacrifice by gross and H&E. Neo-angiogenic markers (Vascular Endothelial Growth Factor, VEGF and αvβ3-integrins) were determined by immunohistochemistry. Doppler indices were compared with the histochemical diagnosis to establish Doppler detection ability of ovarian TAN. Abnormal vascular features including sprouting of blood vessels were suspected by Doppler in 50% of hens and microscopic OVCA were detected in all of these hens. The initiation of decrease in the RI and PI in these hens was recorded from 15–45 weeks. The decrease in Doppler indices was associated with ovarian morphological changes (solid mass). No sustained decrease in Doppler indices was recorded in normal hens. As compared to normal hens, the frequencies of VEGF and αvβ3 expression were greater in hens with decreased DUS indices (RI < 0.4 and PI < 0.5). The results of the study suggest that Doppler ultrasound can detect ovarian tumor associated neo-angiogenesis at early stage of OVCA. This model will establish the foundation of pre-clinical studies with contrast enhanced ultrasound for more precise detection of ovarian TAN and generate valuable information for clinical study." @default.
