- W2022600843 abstract "The aim of this work was to study mating systems in the genus Populus by recording flowering phenology and assessing the frequency of intra- and interspecific mating events using PCR based DNA markers in a range of poplar species growing in Edinburgh in the Royal Botanic Garden and the neighbouring Inverleith Park. The paternity of seedlings from the following three female poplars was studied P. nigra L., P. lasiocarpa and P. maximowiczii. One marker, which could distinguish between P. nigra L. and P. deltoides, was used to look for evidence of introgression from P. deltoides or P.×euramericana into P. nigra L. One microsatellite, developed in P. trichocarpa, and six, developed in P. nigra L., were tested in the 26 hybrid and non-hybrid trees from four taxonomic sections of the genus Populus growing in the Edinburgh collection. There was no evidence of introgression from P. deltoides in the 303 P. nigra L. seedlings that were tested with the species specific marker. It was possible to identify the trees that were acting as the paternal parents in 88% of the 103 P. nigra L. seedlings that were tested with the seven microsatellite markers. The identified male parents were located between 25 and 350 m away from the female P. nigra L. tree. The seedlings produced by the female P. maximowiczii tree that were tested were all fathered by a single P.maximowiczii×P.trichocarpa hybrid tree growing about 175 m away from the female tree. Every seedling that was tested from the female P. lasiocarpa tree was derived by selfing, made possible because this species, unlike most other poplars is monoecious. These results are discussed with regard to the threat which the introduced P.×euramericana and P. nigra L. cv. ‘Italica’ pose to the integrity of the gene pool of P. nigra L. subsp. betulifolia in Britain." @default.
