- W202263176 abstract "The sampling of wild legume populations poses special challenges for plant collectors. Such populations often vary markedly in their density, their age structure, and their flowering and ripening times, making them more difficult to sample than crop plants. In addition, many species either shatter or bury their seeds to aid dispersal or avoid predation. It is important, therefore, that the limited time and resources at the disposal of collectors of these species be as effective as possible in collecting the maximum amount of useful variability. We argue that locally common alleles — alleles with a frequency of >5% in a set of adjacent populations — are the prime target for collectors interested in sampling genes for use directly or in breeding programs.A basic sampling strategy, that maximises the number of locally common alleles collected is (i) to collect 50–100 individuals per site; (ii) to sample individuals at random at each site or microsite visited; (iii) to sample sufficient seeds or other propagates per plant to assure representation of each original plant in all duplicates; (iv) to sample as many sites as possible with 50 per region a notional aim; and (v) to cluster sampling sites in obviously different edaphic and climatic zones so as to sample both the geographic and micro-geographic components of variation. When there is insufficient easily procurable material to collect 50 plants, more sites should be sampled.This basic strategy is recommended when information on the mating system and population genetic structure of the target species is unknown. Where this information is available it is possible to refine the basic strategy to take this information into account. We disagree with recent arguments in the literature that downplay population divergence, and limit total sample size to fewer than 200 per species." @default.
