The uptake of iron by rabbit reticulocytes from the transferrin-iron binary complex and the transferrin-iron-HCO3− ternary complex was studied in order to clarify the role of HCO3− in this process. 2. Only the intact cells were found to be able to detach iron from the ternary complex while from the binary complex iron was also detached by the haemolysate. 3. Q10 values were 14 and 2.5, respectively, when the ternary or the binary complexes were used as substrates. 4. Substituting oxalate for HCO3− in the ternary complex strongly inhibited iron uptake; substitution of malonate produced a weaker inhibition. 5. By using an H14CO3−-labelled ternary complex it could be shown that reticulocytes liberate their HCO3−. 6. Based on these results the following hypothesis is proposed: (after the binding of transferrin to the cell surface) the second step of iron uptake by reticulocytes is the cleavage of HCO3− from the ternary complex. This step is enzymatic and requires an intact cell structure." @default.
