Les differentes familles d’iGluR (recepteurs AMPA, kainate et NMDA) ont une organisation moleculaire semblable, divers domaines d’origine bacterienne ayant ete assembles au cours de l’evolution pour faconner une architecture commune. Durant la derniere decennie, les apports complementaires de la cristallographie et de l’electrophysiologie ont permis des avancees majeures dans la comprehension des mecanismes moleculaires qui regissent l’activation et la modulation des iGluR. Cette revue presente ces mecanismes et montre en quoi une connaissance detaillee du fonctionnement des iGluR permet d’apprehender leur role au niveau de la synapse excitatrice, mais egalement leur potentiel comme cibles d’interet therapeutique dans le traitement de certaines pathologies neurologiques ou psychiatriques." @default.
