- W2023219232 abstract "OBJECTIVE: Apoptosis has been reported in human oocytes and early embryos both in vitro and in vivo. Regulators of apoptosis of the Bcl-2 family are likely to play a determinant role in controlling oocyte and early embryo demise. The aim of the present study is to evaluate the expression of Bcl-2 family genes in the human oocyte, embryos on day 3 (d3) and at blastocyst stage. DESIGN: five human metaphase II oocytes, two d3 embryos and, as representative of the blastocyst stage, eight samples of embryonic stem cells and five trophectoderm cells were collected after informed consent from patients participating in our IVF program. Each sample was individually analyzed by microarray. MATERIALS AND METHODS: After total RNA extraction, amplification and hybridization, all samples were analysed with Affymetrix Genechip human microarrays (HG-U133 Plus 2.0) for gene expression profiling. Data concerning Bcl-2 family genes were analyzed. Statistical comparisons were made with the non-parametric Kruskal-Wallis test. p-values <0.05 were considered to be significant. RESULTS: In mature oocytes, two members are up-regulated: the antiapoptotic Bcl2l10, expressed at the highest level (x 18, p=0.0003), and its proapoptotic homolog Bcl2l13 (x2, p=0,0003). On d3 embryos, Bcl2l10 and Bcl2l13 levels significantly decrease but other members of the family are detected. Indeed, antiapoptotic Mcl1 is significantly up-regulated on d3 embryos (x4, p=0.0004) and persists until the blastocyst stage (x12, p=0.0004). Interestingly, the proapoptotic members Bik and Bim appear to be specifically expressed on d3 embryos (x4, p=0.0145 and x6, p=0.0091 respectively). CONCLUSION: Our study demonstrates that several anti- and pro-apoptotic members of the Bcl-2 family are differentially expressed in the oocyte and during early embryonic development, suggesting specific functions at each developmental stage. The expression of these members will be discussed in relation to oocyte quality and embryo developmental competence." @default.
