- W2023688723 abstract "To investigate the mode of action of Clostridium perfringens epsilon-toxin, MDCK cells were treated with purified toxin and incubated at 37 °C for up to 24 h. Exposure to epsilon-toxin caused a time-dependent decrease in cell–cell and cell–substrate interactions. After 30 min of treatment retraction of the cell body and the emission of filopodia were detectable in a number of cells. Longer exposure resulted in cell rounding and cell blebbing which reached a maximum after 5 h of toxin treatment. A parallel modification in the cytoskeleton was also detected. Actin marginalization and the entanglement of microtubules and intermediate filaments were observed by fluorescence microscopy after 30 min of toxin exposure. Functional alterations of the plasma membrane of MDCK cells were assessed by flow cytometry. After 10 or 30 min of intoxication an increase in cell volume was detected, indicating an alteration in plasma membrane permeability. These findings provide evidence for cytoskeletal changes and plasma membrane functional alterations in the in vitro cell response to C. perfringens epsilon-toxin. Afin d'etudier le mode d'action de la toxine épsilon de Clostridium perfringens, des cellules MDCK ont été traitées avec la toxine purifiée et incubées a 37 °C jusqu'a 24 heures. L'exposition à la toxine épsilon a causé une diminution dependante du temps des interactions cellules–cellules et cellules–substrat. Apres 30 minutes de traitement, la rétraction de la cellule et l'émission de filopodes furent détéctées dans un certain nombre de cellules. Une exposition plus longue a eu pour consequence un arrondissement de la cellule et la formation de gonflements atteignant un maximum apres 5 heures de traitement avec la toxine. En paralléle une modification du squelette cytoplasmique fut aussi detectée. La marginalisation d'actine ét l'enchevetrement des microtubules et des filaments intermédiaires ont été observés par microscopie de fluorescence aprés 30 minutes d'exposition á la toxine. Les changements fontionnels du plasma de la membrane des cellules MDCK ont ét évalués par écoulement cytométrique. Apres 10 ou 30 minutes d'exposition une augmentation du volume de la cellule est observée, indiquant un changement de permeabilité du plasma de la membrane. Ces résultats fournissent l'évidence de l'existence de changements du squelette cytoplasmique et de changements fonctionnels du plasma de la membrane en reponse au traitement in-vitro de cellules par la toxine epsilon du C. perfringens." @default.
