• W2024174856 abstract "Pulmonary and serum antibody responses were evaluated in eight calves vaccinated [four intrapulmonary-right diaphragmatic lobe (IP) and four subcutaneous (SC)] with Pasteurella haemolytica A1 (Ph-1) impregnated agar beads and eight respective sham-vaccinated calves. Experimental and sham groups were challenged in both diaphragmatic lobes with Ph-1 34–37 d after vaccination (DAV) and necropsied 6 d after challenge (DAC; 40–43 DAV). IgG antibodies contained in fluids from the diaphragmatic lobes of vaccinated calves had different patterns of antigen specificity compared with IgG antibodies in analogous sera. Using ELISA, anti-Ph-1 IgA and IgG antibody concentrations were significantly higher (P < 0.05) in lung lavage fluids from the IP group before and after challenge compared to the SC and sham groups. The IP and SC groups developed IgA, IgG and IgM antibody titers in nonvaccinated lung lobes after vaccination and challenge. The IP and SC groups exhibited significantly (P < 0.05) smaller pulmonary lesions than the sham groups and pulmonary IgG and IgA antibodies were associated with increased protection. La réponse immunitaire pulmonaire et sérologique de 8 veaux vaccinés [4 par voie intrapulmonaire (IP) dans le lobe diaphragmatique droit, 4 par voie sous-cutanée (SC)] avec des billes d'agarose imprégnées avec Pasteurella haemolytica A1 (Ph-1) et de 8 veaux témoins ayant reçus un placebo. Les deux groupes de veaux furent inoculés dans les deux lobes diaphragmatiques gauche et droit avec la souche de Ph-1 34–37 jours après vaccination et sacrifiés 6 jours après inoculation, soit 40–43 joirs après vaccination. Anticorps de type IgG présents daus le liquide de lavage des lobes diaphragmatiques reconnaissaient un profil antigénique différent par comparaison aux IgG sériques. Les concentrations en anticorps anti-Ph-1 de type IgA et IgG déterminées étaient significativement plus élevées (P < 0.05) dans les liquides de lavage des poumons pour le groupe de veaux vaccinés par voie intrapulmonaire avant et après inoculation, par comparaison au groupe vacciné par voie sous-cutanée ou avec le placebo. Les 2 groupes de veaux vaccinés développèrent des titres en anticorps de classe IgA, IgG et IgM dans tous les lobes pulmonaires après vaccination et inoculation dans les lobes diaphragmatiques. Les plus hauts titres en anticorps IgA et IgG dans les poumons des veaux vaccinés étaient associés à une protection accrue, correlé par des lésions pulmonaires significativement plus discrètes que dans le groupe témion." @default.
