- W2024210343 abstract "Abstract The mean denaturation temperature or melting point of DNA isolated from freshly ejaculated bovine spermatozoa was 86.43±.25C with an average increase in optical density upon heat denaturation of 37.8±1.3%. The average atomic extinction coefficient for phosphorus was 6,350. Neither storage of spermatozoa for up to 15 days at 4C or 100 days at −79C nor X-irradiation of the spermatozoa followed by storage for up to 8 days prior to extraction of the DNA significantly altered either the denaturation temperature or the increase in optical density from heat denaturation. The melting point of heat denatured spermatozoan DNA was 44.4C. The ionic strength of the medium used to suspend the DNA preparation also influenced the melting profile with a reduction in ionic strength of the medium, shifting the melting point toward lower temperatures." @default.
