- W2024288285 abstract "RATIONALE: Bermuda grass pollen contains a very complexity of allergens, of which only few have been studied. The aim of this study was to characterize Cyn d 24 from Bermuda grass pollen.METHODS: Immunoblot and ELISA were used to study the IgE-binding activity. Purified protein was used to generate mAbs. Recombinant Cyn d 24 was produced from Pichia pastoria. Glycoamidase A and α-mannosidase were used to study the role of carbohydrate moiety in immune response. MALDI-TOF was used to characterize molecular weights of Cyn d 24, enzyme-treated Cyn d 24 and carbohydrate.RESULTS: Cyn d 24 binds only to Con-A indicating the presence of the terminal mannosyl residues. A panel of 5 anti-Cyn d 24 mAbs is mannose-specific and cross-react with proteins from various grass-pollens. Deglycosylated-Cyn d 24 reduced its binding activity to IgE. In addition, α-mannosidase-treated Cyn d 24 reduced its binding activity to IgE.CONCLUSION: Our results indicate that carbohydrate moiety plays an important role in immune response.
