- W2024290613 abstract "The accumulation of endogenous alpha-amylase inhibitor in barley (Hordeum vulgare cv. Bonanza) was studied during grain development. Immature seeds were extracted in either Tris-HCl or Na acetate buffer containing CaCl2 to examine their protein profiles by SDS-PAGE and 2-D gel electrophoresis, as well as for inhibitor accumulation by immunoblotting. The protein profiles showed that there was temporal control over the buffer-extractable proteins. Young seeds contained many species of low abundance proteins that were mostly acidic. As the seeds developed, there was a general shift towards fewer but more abundant proteins. The change in the protein profile was most dramatic during the first 3 weeks of development, and the accumultion of storage-type proteins characterized the later stages. Synthesis of the inhibitor was initiated shortly after fertilization and was most active during early seed development. The protein became one of the more abundant proteins at maturity. A highly specific polyclonal antiserum directed towards the inhibitor was able to detect the inhibitor among many protein species. The inhibitor was present in seed tissues; endosperm and embryo, but not in vegetative tissue under the conditions used in this study." @default.
