- W2024359061 abstract "The immunological maturation of embryonic liver cells was studied using the ability to cause a graft-versus-host (GvH) reaction as an assay. Studies described attempt to determine 1) the direct versus indirect function of liver cells in the GvH reaction, 2) the role of antigen in the maturation of embryonic liver cells, and 3) the role of thymic humoral factors in that maturation.Parental liver cells were injected into thymectomized semiallogeneic F1 irradiated animals whose spleens were subsequently removed and tested for their ability to initiate a GvH reaction in syngeneic or semiallogeneic (F1) test explants. Antisera directed against H-2 of the foreign host component in the test system did not affect the initiation of GvH response by parental liver cells; antisera directed against H-2 of parental liver cells, however, abolished this response.Parental embryonic liver cells were tested for their ability to induce a GvH reaction following the period of exposure to host antigens in the experimental system. Antigens did not seem to affect liver cell maturation in any specific sense because neither partial tolerance nor enhancement of GvH response could be detected following a continuous subjection to F1 antigens.The possible role of thymic humoral function was tested. Syngeneic thymic extracts were injected into thymectomized irradiated recipients along with parental liver cells or were added to the nutrient medium in vitro, While no significant difference was detected between experimental and control groups in the ability of liver cells to cause a GvH reaction when tested 13 or 28 days after thymic extract injection, a difference in initiating the GvH reaction became evident when the assay was performed 4–5 days after injection.From these results it was concluded that: 1) immunological participation of embryonic liver cells in the GvH reaction is specific and direct;?) maturation of liver cells in the host does not appear to be dependent on any specific antigenic stimulation; and 3) thymic extracts can function in liver cell development, apparently by acting on an early phase of this process." @default.
