In addition to succinate, calcium carbonate, MES and phosphate buffers allowed plants to utilize ammonium efficiently. Similar observations were made regarding callus derived from stem tissue of these plants. Plants could be grown in compost, in a physiological state suitable for protoplast isolation, when watered with nutrient solutions containing NH4NO3 and MES buffer. Protoplast division and proliferation to the stage of plant regeneration was possible using Murashige and Skoog (1962) basal medium buffered with succinate, calcium carbonate or MES but not phosphate." @default.
