- W2024790830 abstract "Whole HeLa cells supravitally stained with Janus green B were irradiated with ruby laser light, in order to injure mitochondria selectively. For light microscopic observation, NADH dehydrogenase activity was used as a marker for mitochondria. Immediately after irradiation, the cells stained for NADH dehydrogenase activity showed small vacuoles outlined with the enzyme reaction product and punched-out lesion in cytoplasm, distribution of which corresponded to that of mitochondria in control cells. Electron microscopic observation of irradiated cells disclosed electron opaque particles in mitochondrial matrix and varying sized vacuoles in mitochondria. The light and electron microscopic changes were confined within the mitochondria at the optimum conditions of the dye concentration and the laser light intensity. On morphological bases, it was concluded that mitochondria were selectively injured by the combination of Janus green B supravital stain and ruby laser light irradiation." @default.
