- W2024973812 abstract "Absence, abnormal expression or defective forms of the oocyte activating factor phospholipase C zeta (PLCz) are linked to failed fertilization. This study addressed the effect of motile sperm selection and cryopreservation on PLCz localization. Experimental study. Processed sperm with proven oocyte activation capacity from 14 patients was used either directly for immunostaining or after manually selecting motile sperm (experiment A). Selected motile sperm from 3 patients was stained either fresh or after freezing-thawing (experiment B). An affinity purified antibody specific for full-length PLCz was used. PLCz presence and localization pattern in acrosome intact sperm was determined by laser scanning confocal microscopy. Group comparisons were performed with the Wilcoxon rank test and a P-value of 0.05 was considered statistically significant. A significantly higher percentage of sperm was devoid of PLCz when no selection of motile sperm was performed (20±6% versus 4±1%). The localization pattern of PLCz varied greatly among patients.
