- W2024995901 abstract "Besides the main reactions leading to the repair of tRNA molecules deprived of part or all of their 3′ terminal -pCpCpA sequence, purified E. coli tRNA nucleotidyltransferase catalyzes in vitro, under certain conditions the synthesis of sequences not found in natural tRNAs. In the absence of CTP, AMP is incorporated directly into tRNA-pX or tRNA-pXpC leading to tRNA-pXpA or tRNA-pXpCpA respectively. In the absence of ATP one extra CMP is added to tRNA-pXpCpC to form tRNA-pXpCpCpC. UMP can be incorporated instead of CMP and the sequence -pXpU and -pXpCpU formed. The incorporation of UMP cannot be followed by the incorporation of either a second UMP or an AMP. In all cases, the rate of misincorporation is lower than the rate of the synthesis of the normal sequence. The apparent KM of the enzyme for UTP is 3.0 10−4 M. CTP inhibits competitively the incorporation of UMP into tRNA-pX with a Ki value (1.6 10−5 M) close to its apparent KM. In vitro, outre la réaction principale conduisant à la réparation des molécules de tRNA dont l'extrémité 3′ terminale -pCpCpA est plus ou moins dégradée, la tRNA nucléotidyltransférase de E. coli purifiée catalyse, dans certaines conditions, la synthèse de séquences non observées dans les tRNA naturels. En l'absence de CTP, l'AMP est incorporé directement dans le tRNA-pX ou le tRNA-pXpC, donnant naissance aux tRNA-pXpA et -pXpCpA. En l'absence d'ATP, un résidu CMP supplémentaire est ajouté au tARN-pXpCpC, formant du tRNA-pXpCpCpC. L'UMP peut être incorporé à la place du CMP et les séquences -pXpU et -pXpCpU sont formées. L'incorporation d'un résidu UMP ne peut être suivie de l'incorporation d'un second résidu UMP ou d'un AMP. Dans tous les cas, la vitesse de synthèse de ces séquences anormales est plus faible que celle de la réaction normale. Le KM apparent de l'enzyme à l'égard de l'UTP est de 3.0 10−4 M. Le CTP inhibe compétitivement l'incorporation d'UMP dans le tRNA-pX, avec un Ki (1.6 10−5 M) voisin de son KM apparent." @default.
