- W2025206821 abstract "PURPOSE: Slowing of muscle contraction is a common observation with aging. However, several mechanisms could contribute to this phenomenon, including: reduced calcium ATPase expression and/or activity, reduced expression of calcium binding proteins such as calsequestrin or parvalbumin, or impaired crossbridge cooperativity. In this study, we attempted to identify which, if any mechanims were at work in age-related changes in contractile properties of rat muscle. METHODS: We examined contractile properties, including the twitch half-relaxation time (HRT) and tetanic rate of force relaxation (-df/dt) in the intact plantarflexor muscle group of 8 adult (6-8 months) and 8 old (24 months) f344/BN rats. In addition, we determined sarcoplasmic reticulum (SR) calcium reuptake and expression of calcium ATPase, calsequestrin and parvalbumin in each of the plantarflexor muscles (soleus, plantaris, medial and lateral gastrocnemius). We also determined the myosin:actin ratio in each muscle, as this could theoretically affect crossbridge cooperativity. RESULTS: Twitch HRT were similar in young vs. old muscles (23.1 ± 1.9 vs. 25.9 ± 2.1 ms; P = 0.353). The peak -df/dt was faster in young vs. old muscle (443.7 ± 38.2 vs. 303.5 ± 46.3 mN/ms; P = 0.035). There were main effects of muscle and age (P < 0.001 and P = 0.026, respectively) for calcium uptake, with uptake lowered in old age (∼22%), but the muscle X age interaction was not significant (P = 0.796). The myosin: actin ratio was not altered with age or muscle (P = 0.814 and 0.383, respectively). There were no age-related differences in the expression of calsequestrin, calcium ATPase or parvalbumin. CONCLUSIONS: The maximum rate of tetanic force relaxation, but not twitch HRT, is reduced in the plantarflexor muscles of old rats. Age-associated reductions are also observed in SR calcium uptake, but not in expression of the calcium ATPase or calcium binding proteins. These data are consistent with age-related slowing of force relaxation and calcium uptake being mediated by post-translational modifications." @default.
