CVD 110 a Vc vaccine candidate engineered by deleting the genes encoding cholera toxin (CT), zonula occludens toxin (ZOT), and accessory cholera enterotoxin (ACE), still produces residual small volume diarrhea when fed to healthy volunteers. Using CVD110 supernatant as a negative control in Xenopus leavis oocytes, we incidentally noted a significant swelling of the cells. Based on these observations, this study was designed to examine the effect Of CVD110 supematant on transmembrane sodium transport regulation in isolated rat and rabbit enterocytes. Methods. The Na/K-ATPase activity intracellular accumulation of 22Na, and sodium efflux were determined in enterocytes incubated with var ous dilutions of full supernatant (FSN), and fractionated samples of supematant (FR) with molecular weight pools of 0.5-10, 10-50 and 50-300 kDa. L-broth was used as control in all experiments. Results. CVD 110 supernatant decreased the Na/K-ATPase activity in a dose and time dependent manner. FSN also caused a significant increase in intracellular 22Na accumulation and decrease in 2ZNa efflux, confirming inhibition of sodium pump activity. Of the FR samples, the 10-50kDa fraction caused sigmficant inhibition of the Na/K-ATPase activity and incre-ased intracellular =Na content. The effect on sodium accumulation and Na/K-ATPase activity Of FSN and 10-50kDa fraction are shown in the table ( L-broth=100%; n=4-11 )." @default.
