Minced luteinized rat ovaries were incubated with luteinizing hormone for 1 h, then homogenized and subsequently incubated for 2 h with added luteinizing hormone, [7α-3H]cholesterol and pyridine nucleotides. Control tissues were incubated in the absence of luteinizing hormone or pyridine nucleotides. Luteinizing hormone when present during both the prior incubation and the subsequent incubation with substrate, stimulated the conversion of cholesterol to pregnenolone, progesterone, and 20α-hydroxypregn-4-ene-3-one. The major metabolites of cholesterol in the presence of TPN+ and glucose 6-phosphate were progesterone and 20α-hydroxypregn-4-ene-3-one together with a small amount of pregnenolone. The metabolism of cholesterol by homogenized luteal tissue was negligible in the absence of added pyridine nucleotides.2.2. Luteinizing hormone either given in vivo 3 h previously or added in vitro had no effect on the metabolism of [7α-3H]pregnenolone by minced luteinized ovaries. The major metabolites of pregnenolone were progesterone and 20α-hydroxypregn-4-ene-3-one.3.3. The results indicated that luteinizing hormone stimulated the synthesis of progesterone by an effect on a step between cholesterol and pregnenolone and that the effect was independent of the generation of TPNH." @default.
