The dimerization of bovine mercaptalbumin has been studied in the presence of mercuric salts and of a bifunctional organic mercurial, as a function of pH, temperature, protein concentration, and concentrations of various added ions. Velocity and equilibrium constants have been obtained over a wide range of conditions. The process of dimerization is found to be accurately describable as a second-order process in any given solvent, but the velocity constant varies enormously with the nature of the solvent. 2. Bovine and human mercaptalbumin behave very similarly in general, but the tendency to dimerization is greater in the bovine protein. 3. When bovine mercaptalbumin is dissolved in concentrated urea solutions, light-scattering studies indicate that the albumin molecule is considerably swollen by selective binding of the urea from the mixed solvent. The data are in good agreement with the earlier work of Katz and Doty. No evidence of protein aggregation in urea, apart from the mercury dimerization reaction, was found in our studies. 4. The mercury dimerization reaction takes place readily in ureawater mixtures, the velocity constant being four times as great in 8 M urea as in water at 25 °. The energy of activation is only 12 kcal./mole in 8 M urea, as compared with 18 kcal./mole in water. The amount of dimer formation at equilibrium in 8 M urea is very slightly greater than in water. 5. The effect of pH on dimerization in 8 M urea is qualitatively similar to its effect in water. The velocity constant is a maximum at the isoelectric point in urea solutions, but the corresponding maximum in aqueous solutions is approximately 0.5 pH unit acid to the isoelectric point. Possible interpretations of the data are discussed. 6. The changes produced in bovine mercaptalbumin, by exposure to 8 M urea for 18 hr., are reversible on dilution of the solution to a urea concentration of 1 M, as judged by measurements of light scattering and dimerization." @default.
