In a medium which permits growth, degradation of up to 15 % of the DNA is observed. This degradation does not occur during the post-irradiation time lag, but only during the first doubling of the whole culture. The resumption of growth, characterized by the onset of DNA synthesis, involves every cell, even those which will not form clones. For high doses, which results in very low cell survival, the kinetics follow a different pattern. Degradation begins immediately after ionizing or ultraviolet irradiation, it continues at a lower rate and to a lesser extent than for cultures irradiated at low doses. After ultraviolet irradiation, photoreactivating treatment markedly decreases the degradation observed after low doses. Contrary to the effects of ionizing radiation, degradation after ultraviolet irradiation appears to involve not only the DNA previously synthesized, but also the DNA neosynthesized after irradiation. It may be that, in this situation, the polymerase skips over every “tolerated” dimer, leaving a gap in the newly synthesized DNA strand, whereas after ionizing radiation the break already exists in the irradiated parent DNA." @default.
