- W2027304675 abstract "The colon mucosa plays a fundamental role in sustaining a physical barrier against translocation of pathogenic bacteria and noxious substances from the luminal content of the bowel. 1 Gut ischemia is a common feature in trauma, shock, and bowel strangulation and during repair of abdominal aortic aneurysm and organ transplantation. 2 Reperfusion of the intestinal tract disturbs the barrier function, causing sepsis 3 and ultimately multiple organ failure. A rate-limiting step in the pathophysiology of ischemia/reperfusion (I/R) is the activation and recruitment of leukocytes. 4,5 Activated leukocytes release toxic products, such as oxygen free radicals, proteases, and vasoactive substances, which in turn cause tissue damage and organ dysfunction. 6–11Leukocyte recruitment is a multistep process that is initiated by a rolling adhesive interaction, followed by activation and firm adhesion to the endothelium in postcapillary venules. 12 Leukocyte rolling reduces the velocity and allows time for leukocytes to detect chemotactic substances on the endothelial surface. 13,14 Leukocyte rolling is considered to be predominately supported by the selectin family of adhesion molecules (P-, E-, and L-selectin), although the relative importance of the individual selectins has been reported to differ in specific organs. 4 Firm leukocyte adhesion to endothelial cells is mainly dependent on a group of heterodimeric molecules referred to as β2-integrins, including LFA-1 (CD11a/CD18), Mac-1 (CD11b/CD18), and p150,95 (CD11c/CD18). We and others have recently found that LFA-1 is an important mediator of firm leukocyte adhesion by using CD11a-deficient mice. 15,16 In this context, it is noteworthy that most studies using intravital microscopy to study I/R-induced leukocyte–endothelium interactions in the gastrointestinal tract have mainly been restricted to observations in the small bowel, which is due to a previous lack of models to examine leukocyte responses in the large bowel. Thus, the mechanisms supporting adhesive interactions between leukocytes and the endothelium in the colon are largely unknown. However, a novel approach using inverted intravital fluorescence microscopy (IIVM) has made it possible to analyze the colonic microcirculation;17–19 we have recently developed this approach to study the molecular mechanisms of I/R-provoked leukocyte–endothelium interactions in the colon. 20Based on the considerations above, the purpose of this study was to define the adhesive mechanisms mediating I/R-induced leukocyte rolling and firm adhesion in the colon in vivo." @default.
