- W2028232463 abstract "A single po dose (150–600 mg/kg) of α-naphthylisothiocyanate (ANIT) was administered to rats, mice, and hamsters; dogs received increasingly large doses (50–1000 mg/kg). The hepatotoxic effects produced in these species were compared. Mice were most sensitive; marked hyperbilirubinemia and cholestasis were observed. The response in rats was similar, but higher doses were required for comparable effects. Hamsters were more resistant to the hepatotoxic effects of ANIT. Dogs responded in a different manner; no hyperbilirubinemia was produced even when high doses were administered, although bilirubinuria appeared to be present. In the rodents there was always a parallelism between the periods of cholestasis and hyperbilirubinemia. Hyperbilirubinemia and bilirubinuria in mice have been shown to have a later onset than previously reported, and it is hypothesized that early appearing substances previously described as bilirubin may be metabolites of ANIT capable of undergoing diazotization under certain conditions." @default.
