An apparatus is described in which multiple samples can be fractionated in different pH gradients in less than6 h. Smooth and stable pH gradients were achieved by passing a current of 1 mA through high porosity gels containing 2% (w/v) ampholyte with solutions of 0.01 M H3PO4 and 0.02 M NaOH as anolyte and catholyte respectively. The pH gradients were highly reproducible, and the desired pH range occupied over 80% of the length of the gel. The advantages and limitations of this technique are discussed and examples are given of its application to the separations of several well-characterised proteins." @default.
