- W2028626041 abstract "Orhid-associated bacteria isolated from the underground parts of 12 species of Western Australian orchids were identified by fatty acid analysis and biochemically characterised using a commercially prepared microsubstrate system. Two-thirds of the isolates were successfully identified by the former method. Less than half of the isolates (48%) were Gram-negative, and the most commonly isolated genus was Pseudomonas encompassing 40% of all strains isolated. The next most frequently isolated genera were Bacillus, Kurthia and Arthrobacter, respectively. A small number of isolates belonging to the Enterobacteriaceae were also identified into six different genera. The biochemical features of the main families of bacteria were characteristic, but some isolates within these families had atypical features. The more common bacterial species were isolated from a range of orchids, but some quantitative differences for Bacillus and Pseudomonas spp were evident in the root tissues of Diuris and Pterostylis plants. Approximately 22 and 73% of the bacteria isolated from Diuris and Pterostylis species respectively were Pseudomonas spp. Overall, Gram-negative bacteria were predominant in Pterostylis spp (72.8% of strains) while Gram-positive bacteria were more commonly isolated from Diuris species (71.4% of strains). These differences may be attributed to differences in the underground morphology and physiological status of the plants.
