- W2028916576 abstract "Summary The macromolecular composition (protein, RNA and DNA) of exponential cultures of Halobacterium cutirubrum growing at rates between 0.065 and 0.19 doublings per hour has been determined. The RNA and protein measurements indicate that (i) the cultures are rich in ribosomes, (ii) synthesis of ribosomes is growth rate dependent and (iii) the ribosomes are relatively inefficient, polymerizing only about two amino acids per sec per particle.The DNA content was also high which implies that there may be an unusually high genome copy number. Each genome contains a single copy ribosomal RNA gene cluster which is transcribed as a single 6000 nucleotide long primary transcript containing 16S, alanine tRNA, 23S, 5S and cysteine tRNA sequences. Transcription initiation occurs within each of five conserved octanucleotide direct repeat sequences located in the 900 nucleotide long 5' flanking region. In addition to the octanucleotide sequence, other conserved sequences which might be important in transcription initiation have been identified. Processing of the rRNA primary transcript has been examined by SI nuclease protection assays. Long nearly perfect inverted repeat sequences surrounding the 16S and the 23S rRNA sequences are utilized as initial processing signals to liberate precursor 16S and 23S sequences from the long primary transcript. Mapping evidence suggests that the two transcribed tRNA sequences can either be preserved by endonuclease cutting at or near their mature 5′ and 3′ ends or destroyed by endonuclease cutting within the mature tRNA sequences. The transcription termination signal is probably a tandom GC rich-AT rich sequence beyond the cysteine tRNA gene. Two genes encoding ribosomal proteins HcL20 and HcL11 have been cloned on a 5.5 Kb Clal-BamHI fragment and the clone has been partially characterized by sequence analysis and transcript mapping." @default.
