The biologically active constituent is isolated by solvent fractionation of the acidified concentrate from methanolic extracts, by two successive liquid column chromatography steps and by preparative TLC. The component is detected after TLC (Rf value 0.35, chloroform‐methanol, 88–12) by fluorescence quenching or by staining with vanillin sulfuric acid. Spectral investigations reveal max (methanol) at 228 nm and 264 nm. The retention time on a RP‐18, 5μm LiChroSpher, 4 × 250 mm column (HPLC) is 3.20 min (acetonitril‐water, 45–55). The chemical identity of the component as taxane derivative and its possible mode of action as primarily cytotoxic are discussed. Zusammenfassung Extrakte aus Taxus baccata L. und Taxus baccata cv. fastigiata Loud. hatten Insektizide Wirkung auf Dritt‐ und Viertlarven von Epilachna varivestis . Puppen‐ und Imaginalhäutung wurden gestört. Wirkungen waren nur nach oraler Applikation zu erzielen. Die Aufarbeitung von methanolischen Rohextrakten durch Vorreinigung, 2 Säulenchromatographien und semipräparative Dünnschichtchromatographie führte zur Isolierung der biologisch aktiven Substanz. Der Wirkstoff konnte nach Dünnschichtchromatographie an Kieselgel 60 mit dem Laufmittel Chloroform‐Methanol (88:12) im UV 254 und durch Vanillin‐Schwefelsäure bei einem Rf‐Wert von 0,35 nachgewiesen werden. Sein UV‐Absorptionsspektrum (Methanol) zeigte Maxima bei 228 und 264 nm. Seine Retentionszeit nach HPLC an einer RP‐18 Säule (5 um LiChroSpher, 4 × 250 mm) mit dem Laufmittel Acetonitril‐Wasser (45:55) war 3,20 min. Die Zuordnung der Substanz zur Gruppe der Taxanderivate und ihre Wirkung als Folge cytotoxischer Ereignisse werden diskutiert." @default.
