- W2029458916 abstract "Cyclic 3′,5′-adenosine monophosphate (cyclic AMP) and N6, 2-O′-dibutyryl adenosine monophosphate (dibutyryl cyclic AMP) induced the release of enzyme (chymotrypsinogen and lipase) from rat pancreas in vitro. This release did not exceed 25% of control. Concentration response curves of dibutyryl cylic AMP, carbamylcholine and pancreozymin for secretion have been compared; a similar potency on enzyme release was obtained with dibutyryl cyclic AMP 100 μM, 0.5–1.0 μM carbamylcholine and 0.006–0.008 Ivy unit/ml pancreozymin. The maximal secretion obtained was similar for carbamylcholine and pancreozymin and far lower for dibutyryl cyclic AMP. Theophylline at a concentration of 1.3 mM had no statistically significant effect on the enzyme release from the pancreas but it potentiated a sub-maximal concentration of dibutyryl cyclic AMP. It did not alter the concentratio-response curve of carbamylcholine nor pancreozymin for secretion but occasionally potentiated maximal secretion induced by these agents. Neither cyclic AMP nor dibytyryl cyclic AMP mimicked the metabolic alterations induced by cholinergic agents in the rat pancreas. 0.01 mM carbamylcholine inhibited the incorporation of l-[4,5,-3H2]leucine into pancreatic proteins; dibutyryl cyclic AMP up to a concentration of 2 mM and cyclic AMP (33 mM) had no statistically significant effect on this parameter though being slightly inhibitory at high concentrations. Carbamylcholine (concn. ⩾ 0.005 mM) increased the incorporation of d-[14C6]glucose into pancreatic proteins while dibutyryl cyclic AMP (concn. ⩽ 1.6 mM) had no effect and dibutyryl cyclic AMP (concn. ⩾ 1.6 mM) slightlydecreasedthe incorporation. 0.5 μM carbamylcholine increased the incorporation of 32Pi into the monophosphatidylinositols phospholipid fraction (phospholipid effect). Dibutyryl cyclic AMP up to a concentration of 2 mM did not affect this parameter though it doubled the enzyme release. These results are discussed in three aspects: the involvement of cyclic AMP as mediator of cholinergic agents and pancreozymin, the relationship between stimulation of secretion and the rate of protein synthesis and the relationship of the phospholipid effect with secretion." @default.
