- W2029552077 abstract "Abstract We have carried out circular dichroism studies on bradykinin and an analog of bradykinin in which the prolyl 3 residue has been replaced by 3,4-dehydroproline. The results suggest that the alteration to the prolyl 3 residue exerts an influence on the overall conformation of the analog in aqueous solution. The bradykinin analog containing l -3,4-dehydroproline in the 3-position gives a circular dichroism spectrum in aqueous solution, which, compared to the circular dichroism spectrum of bradykinin in aqueous solution, is consistent with a smaller contribution from a positive Cotton effect in the 220-nm region. In 2,2,2-trifluoroethanol, the circular dichroism spectra of bradykinin and the analog containing l -3,4-dehydroproline 3 more closely approach each other in profile and magnitude. As the pharmacological activities of l -3,4-dehydroprolyl 3 -bradykinin are comparable in kind (23) and in magnitude to those of bradykinin, it is suggested that the similar conformational changes undergone by the peptides with these solvent changes could reflect the conformational changes operative in the hormone-receptor interactions. The circular dichroism changes that are associated with the solvent changes are comparable to dichroism changes that occur with cis trans reorientation around the amide bonds in model l -proline derivatives. As would be expected for these small oligopeptides, the spectroscopic changes are of a low order of magnitude, indicative that the conformational changes are small and probably distributed over a number of possible conformations occurring in solution. It appears reasonable to assume that the conformation of the hormone is subject to the environmental influences of the receptor site. Hence, it is conceivable that the changes produced by organic solvents on the bradykinin conformation could have the same kind of restraint that exists in the receptor binding site." @default.
