- W2030443886 abstract "A sandwich antiglobulin assay was developed in order to characterize the humoral response to weakly immunogenic spontaneous tumors of syngeneic BD X rats. The sandwich technique involving the use of rabbit anti-rat immunoglobulin (Ig) and of radioiodinated protein A was adopted because protein A alone showed low affinity for Ig of the BD X rat strain. Unspecific adsorption of rat and rabbit Ig to plastic surfaces and target cells resulting in an extremely high background response could be eliminated by addition of chicken serum to the test buffers and by absorption of anti-rat Ig on insolubilized normal mouse serum. This was elaborated in model experiments using alloantisera. Sera collected from rats bearing spontaneous tumor transplants showed binding patterns to plated tumor target cells which depended on the tumor line, the route of tumor implantation and the stage of tumor growth. Binding antibodies could be absorbed by tissue cultured tumor cells and fibroblasts, to a lower degree by in vivo grown tumor cells and not at all by erythrocytes and mononuclear cells from spleen and thymus. Sera from untreated Lew and F344 strain rats, which were kept under SPF conditions, contained virtually no antibodies binding to plated tumor cells, but sera from syngeneic BD X rats and from SD rats, both being kept under non-SPF conditions, showed moderate and in some cases exceedingly high binding levels. Evidence is presented to suggest, that at least a portion of these ≪natural≫ antibodies might be directed against Mycoplasma pulmonis, although these microorganisms could not be detected on the target cells. We conclude that the humoral response to weakly immunogenic rat tumors can only be analysed by using Mycoplasma-free rats reared under SPF conditions." @default.
