Die teilgereinigte PEP-Carboxylase aus Bryophyllum und Sedum war durch L-Malat stark hemmbar (K3 ca. 3 mM). Pyruvat vermochte das Enzym aus Bryophyllum stark zu hemmen, war jedoch weniger wirkungsvoll als Inhibitor des aus Sedum angereicherten Enzymes. In Enzym-Rohextrakten aus Bryophyllum war genügend endogenes Malat anwesend, um die PEP-Carboxylase zu hemmen. Fügte man abgekochte Extrakte dem teilgereinigten Enzym hinzu, so ergab sich ebenfalls eine Hemmung, die um so größer war, je mehr Malat in den Extrakten vorlag. Die Aktivitäten der PEP-Carboxylase, Carboxydismutase, malic enzyme und der Malat-dehydrogenase wurden in mit Sephadex gereinigten Extrakten von Bryophyllum und Sedum bestimmt und mit denen anderer Pflanzen verglichen. Die hier vorliegenden Daten stützen die Hypothese, daß eine feed back-Hemmung der PEP-Carboxylase durch L-Malat einer der den Diurnalen Säurerhythmus kontrollierenden Faktoren sein könnte. Phosphoenolpyruvate carboxylase from Bryophyllum tubiflorum has an unusual pH Optimum (pH 6.2–6.5) and extracts of several other succulents with Crassulacean Acid Metabolism show also high activity at pH 6.5. Partially purified phosphoenolpyruvate carboxylase from Bryophyllum and Sedum was inhibited by L-malate (Ki about 3 mM). Pyruvate was a strong inhibitor of this enzyme in Bryophyllum but less effective in Sedum. In crude extracts from Bryophyllum, phosphoenolpyruvate carboxylase was inhibited by endogenous malate. The extent of inhibition of partially purified phosphoenolpyruvate carboxylase by boiled extracts from Bryophyllum was dependent on the malate content of the boiled extract. The activity of phosphoenolpyruvate carboxylase, ribulose diphosphate carboxylase, malic enzyme, and malate dehydrogenase in Sephadex G-25 treated extracts of Bryophyllum and Sedum was compared with activity in other plants. The data support the hypothesis that feed-back inhibition of phosphoenolpyruvate carboxylase by endogenous malate may be involved in the control of Crassulacean acid metabolism." @default.
