- W2031661711 abstract "Orthostatic tolerance is reduced after prolonged head-down tilt (HDT) bedrest. A possible mechanism for reduced orthostatic tolerance could be reduced vasoconstrictor responsiveness. The skin is an accessible vascular bed to assess vasoconstrictor responses, as the effects of local administration of drugs are easily measured. PURPOSE: The objective of this study was to test the hypothesis that 6° HDT bedrest reduces cutaneous vasoconstrictor responsiveness to local norepinephrine (NE) administration, while exercise during bedrest preserves vasoconstrictor responsiveness. METHODS: Twenty subjects (18 males and 2 females) underwent 14 days of strict 6° HDT bedrest. Thirteen of these subjects exercised for 90 min a day at 75% of pre-bedrest heart rate maximum, while 7 subjects did not exercise during bedrest. Intradermal microdialysis was used to locally administer 8 doses of NE (range from 1•10−11 through 1•10−2 log-M). NE doses were infused for 5 min each at a rate of 2 μliters per min. Forearm skin blood flow (SkBF; laser-Doppler flowmetry) was monitored directly over the microdialysis membranes. Nonlinear logistic modeling was used to identify EC50 (i.e., concentration resulting in half of the maximum vasoconstrictor response), minimum, and maximum values from the SkBF/NE dose response curve. RESULTS: HDT bedrest did not significantly affect the EC50 (−6.0 ± 0.2 to −5.6 ± 0.4 log-M) or minimun and maximum SkBF. Likewise, the EC50 (−5.5 ± 0.2 to −5.4 ± 0.2 log-M), minimum, and maximum SkBF were not significantly different after bedrest in the exercise group. Finally, no significant interaction was observed between main factors (i.e., pre/post bedrest and exercise/non-exercise condtions) for any variable. CONCLUSION: These data suggest that 14 days of HDT bedrest, with or without exercise, do not alter cutaneous vasoconstrictor responses to exogenous NE.
