- W2034152581 abstract "The circular dichroism spectra of amino acid specific t-RNA in the presence of magnesium show four distinct steps as a function of temperature. Of particular interest is the sudden increase of the CD spectrum between 36 and 40°C. This increase is absent in the absence of magnesium and in charged t-RNA. From the melting curves a van t'Hoff plot can be constructed and thermodynamic parameters can be estimated. This plot permits the separation of the two last steps which are distinct in their thermodynamic parameters; these indicate that t-RNA behaves rather like a globular protein than a polynucleotide. Le spectre de dichroïsme circulaire du t-RNA spécifique d'un amino-acide en présence de magnésium présente quatre paliers différents en fonction de la température. On notera en particulier l'augmentation brusque du spectre de dichroïsme circulaire entre 36 et 40°C. Cette augmentation n'intervient pas en l'absence de magnésium et dans le t-RNA chargé. A partir des courbes de fusion on peut établir un diagramme de van t'Hoff et définir les paramètres thermodynamiques. Ce diagramme permet la séparation des deux derniers paliers qui présentent des paramètres thermodynamiques distincts. Ceci montre que le t-RNA se comporte plus comme une protéine globulaire que comme un polynucléotide. Aus den Schmelzkurven kann eine graphische van t'Hoff-Darstellung konstruiert und thermodynamische Parameter können geschätzt werden. Diese Darstellung erlaubte die Trennung der beiden letzten Etappen, die durch ihre thermodynamischen Parameter verschieden sind : diese zeigen an, dass die t-RNA sich mehr wie globulare Proteine als wie Polynukleotide verhalten." @default.
