- W2034188039 abstract "This chapter describes the methods for the assay of catecholamine-synthesizing enzymes. Catecholamines, such as dopamine, noradrenalin, and adrenaline are synthesized from tyrosine. Four enzymes are involved in the biosynthesis of adrenaline: (1) tyrosine hydroxylase (TH), (2) aromatic L-amino acid decarboxylase, (3) dopamine β-hydroxylase, and (4) phenylethanolamine N-methyltransferase. TH, a pteridine-requiring monooxygenase, catalyzes the first step in the biosynthesis of catecholamines. TH requires both L-tyrosine and molecular oxygen as substrates and a tetrahydropteridin (2-amino-4-hydroxy-5,6,7,8-tetrahydropteridin) as co-factor, producing DOPA, H2O, and a quinonoid dihydropterin as products. TH is localized in the adrenergic and noradrenergic adrenal medulla cells, as well as dopaminergic, noradrenergic, and adrenergic nerve cells. The chapter describes the methods for determination of tyrosine hydroxylase activity and methods for determination of tyrosine hydroxylase protein by a sandwich enzyme immunoassay. It also discusses the assay methods for aromatic L-amino acid decarboxylase activity and assay methods for dopamine-p-hydroxylase acitvity. It also presents assay methods for phenylethanolamine JV-methyltransferase activity." @default.
