- W2034651057 abstract "Twenty diploid clones of Citrus species and hybrids and one diploid clone of Fortunella were treated with colchicine to develop polyploids for use in breeding experiments. Details on the techniques used for plant preparation, colchicine treatment, identification and isolation of polyploid tissue, and the microtechnique used to identify the cytochimeras produced are described. Diagnostic features found to be useful in macroscopic screening were sectorially patterned leaves, leaf color changes, petiole blade shape, thickened leaf venation, comparative size differences in oil glands and stomates, leaf blade thickness, and leaf shape. Eight types of cytochimeras were identified from cytological examination of colchicine-affected material. Data are given for cultivars polyploidized by colchicine, type of histogenic layer change isolated per clone, frequency of isolations per clone, and certain leaf measurements and relationships. Data are also presented for the number and frequency of layer changes and distribution and frequency of types of cytochimeras in polyembryonic and monoembryonic clones. Results demonstrate that in the induction and expression of polyploidy, a differential response to colchicine exists in the genetic constitution of clones, in the individual histogenic layers, and in the presence or absence of adventitious embryony The frequency of cytological types and layer associations is discussed in relation to possible modes of origin. Aspects of layer association and origin in relation to intraclonal variation and citrus breeding are discussed Some potential uses of citrus cytochimeras are mentioned." @default.
