Our first aim was to test the hypothesis that accumulation of electrons in cytosolic free NADH, the reduced form of nicotinamide adenine dinucleotide (NAD), functions as this sensor. The rationale for testing this hypothesis was based on the near equilibria between extracellular and intracellular lactate/pyruvate (L/P) and free cytosolic NADH/NAD+ ratios; NADH was increased or decreased by injecting lactate or pyruvate, respectively. Our second aim was to identify components of the signaling cascade that mediate increased flows.PRINCIPAL FINDINGS1.
