- W2037229853 abstract "A new extended-release formulation of cefaclor, a second-generation oral cephalosporin, was developed to permit twice-daily (BID) dosing. When compared with immediate-release cefaclor, which is administered three times daily (TID), the extended-release formulation appears to enhance dosing convenience and thereby improve patient compliance and clinical outcome. Double-masked, randomized clinical trials involving more than 5000 patients were conducted in the United States and Europe to assess the relative efficacy and tolerability of different doses of extended-release cefaclor and immediate-release cefaclor in the outpatient treatment of several infections caused by susceptible pathogens. Data from most of these trials have been published previously, whereas other data are reported here for the first time. The results indicate that extended-release cefaclor administered in dosages of 750 mg BID for 10 days in acute bacterial sinusitis; 375 mg BID for 10 days in pharyngitis/tonsillitis; 375 mg, 500 mg, or 750 mg BID for 7 days in acute bronchitis and acute bacterial exacerbations of chronic bronchitis; 500 mg once daily or 375 mg BID for 7 days in uncomplicated cystitis or asymptomatic bacteriuria; 500 mg BID for 14 days in pyelonephritis; and 375 mg BID for 7 days in skin and skin-structure infections appears to be as effective and well tolerated as immediate-release cefaclor administered in clinically comparable doses for equivalent TID time periods" @default.
