- W2037293868 abstract "Background: Arachidonic acid (AA) is a key biological intermediate that is converted to a large number of eicosanoids with potent biological activity. AA is metabolized through two major pathways namely cyclooxygenase (COX) and lipoxygenase (LOX) pathways. The COX pathway leads to formation of prostaglandins and thromboxanes, while the LOX pathway leads to formation of leukotrienes. Prostaglandins are known to cause acute inflammation. Leukotrienes are powerful chemotactic agents for both neutrophils and macrophages. Both COX and LOX enzymes individually have been targeted for containing inflammation. The COX inhibition has been associated with shunting of AA to the LOX pathway. Pharmacological inhibition or gene deletion studies on COX-2 and 5-LOX have shown a protective effect in experimental models of acute pancreatitis. However, there are no studies to indicate, whether simultaneous inhibition of both pathways is more protective. Therefore, the aim of the present study was to evaluate the effect of blocking both COX-2 and 5-LOX enzymes simultaneously on the severity of acute pancreatitis. Methods: We studied the effect of caffeic acid (70 mg/kg, two injections), a lipoxygenase inhibitor on caerulein induced pancreatitis in COX-2 knock out and wild type mice. Severity of pancreatitis was evaluated by studying changes in serum amylase, pancreatic myeloperoxidase and histopathology. Results: Caffeic acid administration decreased plasma amylase, myeloperoxidase and histopathological changes in both wild type and COX-2 knockout mice as compared to untreated controls. However, treatment of COX-2 knockout animals with the lipoxygenase inhibitor did not further enhance the protective effect as compared to wild type treated with lipoxygenase inhibitor. Conclusion: Total inhibition of AA metabolites through dual inhibition of COX-2 and 5-lipoxygenase does not provide additional protection from injury in caerulein-induced pancreatitis compared to individual inhibition." @default.
