- W2037350430 abstract "Polyvinyl sulphate (PVS) was found to induce a dissociation of purified 80-S ribosomes from plant leaves, yeast and rabbit reticulocytes into subunits. The dissociation occurred both at room and low temperatures with concentrations of PVS of 0.4–10 mg/ml. Dialysis of PVS against the Mg 2+ containing buffers used or even against 0.1 M MgSO 4 before use did not in any way change its ability to bring about the dissociation which, therefore, seems to differ from that induced by the removal of Mg 2+ from the particles. It was also shown that incubation of the ribosomes with 4 mg/ml PVS for periods of up to 3 h at 4° did not produce any detectable change in the sedimentation characteristics of the RNA isolated from them. These results suggest that some basic component in addition to Mg 2+ may be important to the stability of 80-S ribosomes. The subunits formed from the ribosomes of different origins were all found to have s 20,w values of about 50 S and 34 S. Ribosomes from plant leaves and reticulocytes but not those from yeast yielded in addition 2 faster-moving components with s 20,w values of about 70 S and 100 S. The proportion of the material present as these faster-moving components was increased by a higher ratio of Mg 2+ to phosphate in the solution but was unaffected by the ribosome concentration over the range 0.5–6 mg/ml." @default.
