- W2037392392 abstract "To examine whether sperm apoptosis correlated with abnormal sperm head morphology using IMSI technique of magnification and conventional staining. 78 consecutive semen samples exhibiting teratospermia were evaluated for head defects by IMSI morphology and conventional staining and sperm apoptosis. Inclusion was presence of teratospermia as per Kruger's criteria. Sperm head defects were compared by conventional staining by Papanicolaou technique and IMSI morphology. Sperm apoptosis testing for DNA fragmentation was performed by the TUNEL technique. Sperm apoptosis was classified normal (13%) (Group A), borderline (14-40%) (Group B), and abnormal (>40%) (Group C). 42/78 (53.8%) samples in Group A, showed normal sperm apoptosis and on an average 67.4% sperm showed head abnormalities by IMSI technique and 76.9% by conventional staining. 30/78 (38.5 %) samples in Group B, showed borderline apoptosis. In this group on an average, 68.4% sperm showed head abnormalities by IMSI technique and 76.6% by conventional staining. 6/78 (7.7%) samples in Group C, showed abnormal apoptosis. In this group, on an average 66.8% sperm showed head abnormalities by IMSI technique and 68.7% sperm by conventional staining. The head morphology abnormality did not worsen with increase in DNA fragmentation. Also, there was no significant difference between the results of IMSI and conventional staining.Tabled 1IMSI vs CASA vs Sperm apoptosisIMSI (total sperm)CASA (total sperm)Head AbnormalityHead AbnormalitySperm ApoptosisAbnormal %Abnormal %IMSI %CASA %Average (%)Group A (n=42)93.592.967.476.98.7Group B (n=30)93.792.968.476.622.8Group C (n=6)95.094.566.868.765.8 Open table in a new tab This study suggests that there is no correlation between increased sperm DNA fragmentation and abnormal sperm head morphology. Also, conventional staining can itself detect head abnormalities and hence morphology testing using IMSI technology may not be necessary to determine sperm head abnormalities." @default.
