- W2038518272 abstract "We investigated the fate of the major outer membrane protein of Neisseria gonorrhoeae, P.IA, during gonococcal infection of Chang conjunctiva epithelial cells by using immunoelectron microscopy. Probing of P.IA epitopes with mono- and polyclonal antibodies revealed variable, fixation-dependent P.IA epitope exposure in the gonococci used as an inoculum in the infection experiments. Detection of invariable exposed P.IA epitopes in cryosections of infected epithelial cells with a polyclonal antiserum revealed unaltered P.IA exposure on the bacterial membranes during early attachment of the bacteria to the eukaryotic cells. Upon entry of the bacteria into the host cells, however, labelling was extended to membraneous structures that intercalated between the bacteria and the host cell surface, and, occasionally, to the host cell plasma membrane. The latter observation is consistent with the suggested active role of P.I in the uptake process (as shown in 1985 by E.C. Gotschlich). Once inside the epithelial cells, both morphologically intact and disintegrating bacteria could be distinguished. The disintegration of the bacteria was accompanied by a loss of P.IA immunoreactivity. Nous avons observé en microscopie immunoélectronique le devenir de la P.IA de Neisseria gonorrhoeae au cours de l'infection gonococcique des cellules épithéliales conjonctivales (lignée Chang). L'étude des épitopes au moyen d'anticorps monoclonaux et polyclonaux a montrée la présence variable de l'épitope P.IA en fonction de la fixation, chez les gonocoques utilisés comme inoculum dans l'infection expérimentale. La détection d'épitopes invariablement présents, à l'aide de cryosections de cellules épithéliales infectées et d'immunsérums polyclonaux, a mis en évidence une présentation immuable de la P.IA sur la membrane bactérienne au cours de la fixation précoce de la bactérie sur la cellule eukaryote. Cependant, la pénétration bactérienne dans la cellule hôte est caractérisée par une extension du marquage vers les structures membranaires, à la jonction des surfaces des bactéries et des cellules hôtes et parfois jusqu'à la membrane cytoplasmique de ces cellules. Cette dernière observation est conforme au rôle actif de la P.I suggéré (selon Gotschlich en 1985) dans le processus d'absorption. Dès lors que les bactéries se trouvent à l'intérieur des cellules épithéliales, on reconnaît à la fois des bactéries morphologiquement intactes et des bactéries qui se désintègrent. La désintégration bactérienne s'accompagne d'une perte de l'immunoréactivité de la P.IA." @default.
