- W2038587975 abstract "Abstract The effect of a Phenoclor DP6 diet on the time course of induction of liver microsomal enzymes was studied in young rats. In experiment A, rats were fed a diet containing 10 ppm of DP6 on only Day 1 of the experiment. In experiment B, rats were fed a DP6 diet during the whole experiment. Rats were killed 1, 3, 5, and 8 days after the beginning of the DP6 treatment. [ 14 C]Arginine was given ip before sacrifice. The microsomal fraction of the liver was prepared and analyzed for aniline hydroxylase and aminopyrine N -demethylase activities, cytochrome P -450 and cytochrome b 5 contents and specific protein radioactivity. In experiment A (DP6 ingested: 2 mg/kg) all the microsomal indices studied were increased by DP6 treatment except for cytochrome b 5 . This enhancement was maximal in 5 days and afterward the activities began to decline. The time-dependent profile of incorporation of [ 14 C]arginine into microsomal proteins corresponded well with those of the enzyme activities. In experiment B (DP6 ingested: 2 mg/kg/day) the induction patterns of cytochrome concentrations and enzyme activities were different: For the monooxygenases, the maximal activities were observed 3–5 days following the beginning of DP6 ingestion and then remained significantly higher than control values; for the cytochromes, the microsomal concentrations (including inactive forms) continued to increase the entire 8 days of treatment. The time profile of the incorporation of [ 14 C]arginine into microsomal proteins was similar with those of monooxygenase enzymes. The increased cytochrome content of microsomes did not seem to be dependent on increased protein synthesis. The induction profile of microsomal indices seems to be dependent upon the nature of the PCB compounds and their selective metabolism." @default.
