Experiments have been carried out in order to compare the efficiencies of colchicine, p-dichlorobenzene, α-bromonaphthalene, acenaphthene, and 8-hydroxyquinoline for accumulating metaphases in Vicia faba root meristem cells. 2. It was found that colchicine was superior to the other spindle inhibitors tested and that roots treated for short periods in non-aerated colchicine solutions had a reduced metaphase accumulation as compared with roots treated in aerated solutions. The decrease in metaphase accumulation in non-aerated colchicine solutions was shown to be the result of a decrease in the rate of entry of cells into mitosis under anoxic conditions. 3. In experiments with p-dichlorobenzene it was found that roots treated in non-aerated solutions and in deoxygenated solutions, showed a significant increase in metaphase accumulation over roots maintained in fully aerated solutions. 4. The difference in the response between roots treated with p-dichlorobenzene and colchicine in the presence or absence of oxygen, would appear to be a reflection on the differences in the mode of action of these substances. It is suggested that the enhanced action of p-dichlorobenzene in the absence of oxygen may be due to a decrease in the cells capability for detoxicating this substance." @default.
