- W2039116875 abstract "The synthesis and degradation of collagen by bones with high (0.107 per cent), moderate (0.041 per cent) and low (0.007 per cent) fluoride contents were assessed in organ culture, using half-calvaria of 5-day-old mice. Fluoride was incorporated into the half-calvaria before culture, during the entire period of bone formation. Collagen synthesis was measured by the conversion of [14C]-proline to [14C]-hydroxyproline, and collagen degradation by the release of hydroxyproline from bone. Fluoride inhibited the loss of hydroxyproline, both in control cultures and in the presence of parathyroid hormone. Calcitonin inhibited the release of hydroxyproline, and fluoride further reduced the loss from calcitonin-treated bones. Fluoride did not influence the conversion of [14C]-proline to [14C]-hydroxyproline, but enhanced the incorporation of newlysynthesized collagen into bone matrix, and reduced its subsequent loss by resorption. Parathyroid hormone suppressed collagen synthesis, and reduced the incorporation of [14C]-hydroxyproline into bone matrix. La synthèse et la dégradation du collagène dans l'os à contenu élevé (0,107 pour cent), modéré (0,041 pour cent) et faible (0,007 pour cent) en fluor ont étéétudiées en cultures de tissu, en utilisant des moitiés de calottes craniennes de souris âgés de 5 jours. Le fluor est incorporé dans l'os avant la mise en culture pendant la période totale de formation osseuse. La synthèse collagénique est estimée par le changement de [14C]-proline en [14C]-hydroxyproline et la dégradation collagénique est appréciée par la libération d'hydroxyproline de l'os, en cultures de tissu et en présence d'hormone parathyroidienne. La calcitonine inhibe la libération d'hydroxyproline de l'os; cette inhibition peut être augmentée par le fluor au niveau de l'os traité par calcitonine. Le fluor n'intervient pas dans la transformation entre [14C]-proline et [14C]-hydroxyproline, mais augmente l'incorporation de collagène nouvellement synthétisé dans la matrice osseuse et réduit sa destruction par résorption. La parathormone empêche la synthèse du collagène et réduit l'incorporation de [14C]-hydroxyproline dans la matrice osseuse. Synthese und Abbau von Kollagen in Knochen mit hohem (0,107 Prozent), mittlerem (0,041 Prozent) und geringem (0,007 Prozent) Fluoridgehalt wurden unter Verwendung von halbierten Calvarien 5 Tage alter Mäuse in Organkulturen geprüft. Fluorid wurde während der gesamten Knochenbildungsperiode vor Beginn der Kultur in die Halbcalvarien inkorporiert. Die Kollagensynthese wurde durch Umbau von [14C]-Prolin in [14C]-Hydroxyprolin gemessen, der Kollagenabbau durch Freisetzung von Hydroxyprolin aus Knochen sowohl in den Vergleichskulturen als auch unter Einfluβ von Parathormon. Calcitonin hemmte die Freisetzung von Hydroxyprolin, und Fluorid verminderte weiterhin den Verlust von Hydroxyprolin aus Calcitoninbehandelten Knochen. Fluorid beeinfluβte nicht die Umwandlung von [14C]-Prolin zu [14C]-Hydroxyprolin, es steigerte jedoch die Inkorporierung frisch synthetisierten Kollagens in die Knochenmatrix und verminderte zugleich dessen Verlust durch Resorption. Parathormon drückte die Kollagensynthese herab und verminderte die Inkorporierung von [14C]-Hydroxyprolin in die Knochenmatrix." @default.
