- W2039264906 abstract "Photosynthetic O2 evolution by 1 mm wide slices of leaves from young barley seedlings (Hordeum vulgare L.) was measured under varied conditions using manometric and polarographic methods. Effects of pH and carbonic anhydrase affecting the relative availability of CO2 and HCO3- in the experimental solutions show that only CO2 but not HCO3- is utilized in photosynthesis of the leaf slices. The maximum rates of O2 evolution obtained with 1 mm leaf slices (250 to 350 µmol O2 • g-1 fresh wt • h-1 or 150 to 220 µmol O2 • mg-1 chlorophyll • h-1) were only 15 to 20% lower than the rates obtained with larger (25 mm) unsliced leaf segments. CO2 was taken up predominantly through the stomata and only to a limited extent by diffusion via the cut edges. Thus when leaf slices or larger leaf segments are submerged in experimental solutions stomata function in gas exchange in a similar way as in the intact plant. In experiments with leaf slices in experimental solution, a material widely used in investigations of active, photosynthesis-dependent ion uptake, it is essential to maintain conditions which guarantee a sufficient CO2 supply to obtain maximum rates of photosynthesis." @default.
