- W2039777225 abstract "Abstract The uptake and the biosynthesis of 5-HT was studied in the isolated superior cervical ganglion of the rat. The ganglia of normal animals do not contain detectable amounts of 5-HT. They possess, however, a very active 5-HTP decarboxylase. 5-HT evolved by decarboxylation of 5-HTP in the isolated ganglia is catabolized by monoamine-oxidase. The degradation may be suppressed by pretreating the experimental animals with pargyline, a monoamine-oxidase inhibitor. In isolated ganglia, obtained from pargyline pretreated rats, 5-HT evolved within the ganglia diffuses into the bathing fluid, and thus escapes destruction by monoamine-oxidase. 5-HT concentrations in ganglia incubated in Krebs' solution gassed with 95% N2 + 5% CO2 (instead of 95% O2 + 5% CO2) were much higher than under aerobic conditions, when either 5-HT or 5-HTP were added to the incubation medium. The absence of 5-HT in the living superior cervical ganglion, which possesses an active 5-HTP decarboxylase, cannot be explained solely by the action of monoamine-oxidase, since ganglia of rats pretreated with pargyline do not contain detectable amounts of 5-HT. The increase in intraganglionic 5-HTP concentration under anaerobic conditions could be due to the presence of an oxygen-dependent degrading system other than monoamine-oxidase. No such system is actually known to exist in the superior cervical ganglion. A more probable explanation for the maintenance of an electro-chemical 5-HT gradient across the ganglion membrane, which could be responsible for the absence of detectable amounts of 5-HT in the living ganglion, is the presence of an active outward transport system of 5-HT." @default.
