- W2040055802 abstract "Summary Experiments on the suitability of the weak acid 5,5-dimethyl-[2- 14 C]-oxazolidine-2,4-dione (DMO) as a chemical probe for the determination of cytoplasmic pH in leaves of higher plants have been performed. The non-dissociated acid (H-DMO) is the major species taken up passively by leaf slices incubated in 14 C-DMO. However, a stable steady-state distribution of 4 C-DMO between the tissue and the incubation medium is not reached. Together with other problems (e.g. DMO-metabolism in the tissue, finite permeability for the anion DMO - , compartmentation, secondary changes during incubation of leaf slices) this prevents calculation of absolute values of cytoplasmic pH. Nevertheless, the initial rate of 14 C-DMO uptake, which is constant for incubation of 20 to 40 min, is proportional to pH of the bulk cytoplasm. It is thus possible to study relative changes of cytoplasmic pH in leaf cells in relation to crassulacean acid metabolism (CAM). Characteristic changes were observed in leaf or phyllodium cells of the CAM plants Kalanchoe daigremontiana, K. tubiflora and K. blossfeldiana cv. Tom Thumb in the light and dark period. It is concluded that these changes, which presumably are of small magnitude, are a reflection of metabolic control of cytoplasmic pH during CAM." @default.
